Application of a Small Protein-Coated Column to Trap, Extract and Enrich Carbamazepine Directly from Human Serum for Direct Chromatographic Analysis

An automated solid phase extraction (SPE) protocol to determine carbamazepine in human serum has been developed and validated using a simple, rabid sensitive liquid chromatography-based bio-analytical method. Extraction of was carried out an on-line SPE tool short protein-coated (PC) ODS silica pre-column (PC-ODS-pre-column) phosphate buffer saline (PBS) with pH 7.4 as solvent. There are two distinct chromatographic modes used by PC-ODS-pre-column. While trapping required reversed-phase chromatography, proteins were extracted from samples PBS size-exclusion chromatography. Then, eluted the PC-ODS-pre-column onto quantification position mixture methanol-distilled deionized water (50:50, v/v) eluent analytical column. At room temperature (22 ± 1 °C), completely separated co-eluted matrix components detected at 230 nm. Carbamazepine’s linearity obtained concentrations ranging 50 10,000 ng/mL. With good accuracy precision, recoveries ranged 86.14 97.82%. The step conducted safe green solvent, making this both cost-effective ecologically safe.
